Bring to room temperature and strip
foil from required number of wells.
Empty wells and tap dry onto tissue.
Add 10µl of standard or sample
to each well.
Add 200µl progesterone-enzyme
label.
Leave 1-1.5 hours at room
temperature.
Wash 3 times with cold water
- tap dry on paper each time.

Add 200µl substrate in
substrate buffer. To avoid
contamination use a clean tip
Leave 10 min - 1/2 hour.
Compare colour development of
samples with standards by eye.
Strong colour - low progesterone ?
heat, not pregnant
Weak colour - high progesterone ?
mid cycle or pregnant

alternative substrate Sigma 104 at
405nm: 140mg/25ml buffer).
Samples
Whole bovine milk. May be stored at
4°C after addition of Lactab MkIII
tablets.
Standards
Progesterone in milk from an oestrus
or ovariectomized animal.
WARNING
All solutions contain thimerosal
(0.01% w/v).This is toxic. Dispose
of carefully and wash down sinks
with excess water. Keep away from
food and children and wash your
hands after use.
*Dissolve
substrate tablets in 25ml substrate
buffer 2 and mix.
Takes 20-30 minutes to dissolve.
Sufficient for one plate. Store in
the dark.
Do not contaminate with
progesterone-enzyme label 1. |
